PROFIL FARMAKOKINETIKA NATRIUM DIKLOFENAK SETELAH PEMBERIAN JUS SAWI HIJAU (Brassica rapa var. parachinensis L.) PADA TIKUS PUTIH JANTAN GALUR WISTAR

Induction of the enzyme is an important rule that has found implications in pharmacology and toxicology of a drug. Due to the induction of  biotransformation enzymes, the rate as will increase. Mustard green (Brassica rapa var. parachinensis L.) is one of vegetable which is  liked by the people of Indonesia. The content of indole in the mustard green can act as enzyme in the biotransformation of diclofenac. So it can decrease the number of active diclofenac inflammatory therapy. this research is an experiment using 21 white male rats of  wistar strain fasted for 12 hours divided into 3 groups each group contains 7 animals,than in their blood taken through orbital vein at  0, 0.5, 0.75, 1, 1.5, 2, 3, 4, 6, 8, 11 hours. Determination of diclofenac sodium performed with UV spectrophotometer at wavelength of 278 nm. Calculated pharmacokinetic parameters are (Ke, t ½, Ka, tmaks, AUC, Cpmaks, Vd, Cl). The parameters are calculated based on a compartment and analyzed statistically by anova test using SPSS version 13:00. The results showed the provision of mustrad green juice for 1 day with a dose of 35.28 g / kg weight for rats and 7 days with a dose of 5.04 g / kg weight for rats before given diclofenac sodium, the Clereance of diclofenac sodium can increase by 24.3479%, and 33, 38% (P <0.05) and  Cpmaks decreased by 50.90% and 68.57% (P> 0.05).

AKTIFITAS HIPOKOLESTEROL AIR REBUSAN JAMUR KUPING HITAM (Auricularia polytricha (Month.) Sacc.) PADA TIKUS YANG DIBERI DIET LEMAK TINGGI

The aim of this research was to know the effects and dosage of Jamur Kuping Hitam (Auricularia polytricha (Month.) Sacc. ) water extract as hipocholesterol. The research was done with post test only randomized control group design. Animal trials Wistar male rats as much as 25 divided by 5 groups. Group I (negative control) were given aquadest, II (positive control) were given Simvastatin 1,26 mg/kg, groups III, IV and V were given Jamur Kuping Hitam water extract 6,3; 12,6 and 25,2 ml/kg. All groups were given a high fat diet (DLT) which is mixtures standard feed and oil pigs (90:10) for 7 days. All treatment was done by oral. Measurement of total cholesterol concentration done on days 7 and 14 after treatment. Therefore, total cholesterol concentration was analyzed statistically with the Kruskal-Wallis and Mann-Whitney using SPSS release 16. The results obtained Jamur Kuping Hitam (Auricularia polytricha (Month.) Sacc.) water extract have effect hipocholesterol with dosage is 25,2 ml/kg.

Isolation and Identification of Antimicrobial Compound From Sarang Semut Tubers (Myrmecodia tuberosa (non Jack.)Bl.)

Umbi Sarang Semut (Myrmecodia tuberosa (non Jack.)Bl.) merupakan tanaman obat tradisional yang banyak digunakan untuk melawan berbagai penyakit di Papua Barat. Akhir-akhir ini telah diketahui bahwa ekstrak umbi sarang semut mempunyai senyawa aktif yang dapat melawan kerja antibakteri.

Ekstrak kental etanol umbi sarang semut didapatkan dengan metode maserasi menggunakan etanol 95%. Ekstrak kental etanol yang didapatkan kemudian difraksinasi dengan n-hexana, etil asetat, dan air. Ekstrak kental dan fraksi yang didapatkan diuji aktivitas antibakterinya dengan menggunakan bakteri Staphylococcus aureus (0,890 cm). Karakteristik senyawa dari fraksi etil asetat memberikan hasil bahwa senyawa yang mempunyai aktivitas antibakteri terhadap bakteri Staphylococcus aureus merupakan senyawa fenolik.

MOLECULAR DOCKING, SINTESIS DAN UJI AKTIVITAS SITOTOKSIK SENYAWA 1-(3-KLOROBENZOIL)-1,3-DIMETILUREA

A novel 1-(3-chlorobenzoyl)-1,3-dimethylurea has been designed, synthesized, structurally determined, and the in vitro cytotoxic activity was evaluated. Docking simulation was performed to the position of this compound into the Checkpoint kinase 1 (Chk1) active site to determine the probable binding model. Synthesis of 1-(3-chlorobenzoyl)-1,3-dimethylurea was completed by acylation reaction between 1,3-dimethylurea and 3-chlorobenzoyl chloride. The purity of synthesized product was determined by Thin Layer Chromatography and melting point measurement. Structure identification was performed by UV and FTIR spectrophotometer, NMR spectrometer. Antiproliferative assay result demonstrated that this compound possessed good cytotoxic activity against HeLa cells, which is comparable to the positive control. This compound with potent cytotoxic activity might be a potential anticancer agent.

PENGARUH JUS BUAH MANGGIS (Garcinia mangostana L.) TERHADAP PROFIL FARMAKOKINETIK PARASETAMOL TIKUS PUTIH JANTAN (Rattus norvegicus) GALUR WISTAR

Drug interaction happens when drugs is given together with foods. This research was aimed to observe the effect of mangosteen juice to the paracetamol pharmacokinetic profile in Wistar male white rats. The study used Wistar rats weight of 200-350g. The animals were divided into three groups with the total of the male rats are 15. Group I (paracetamol) was given a single oral paracetamol 9 mg/200gBW. Group II (treatment 1) was given a single oral paracetamol 9 mg/200gBW together with mangosteen juice 2,88 g/200gBW. Group III(treatment 2) was given a single oral paracetamol 9 mg/200gBW together with mangosteen juice 0,66 g/200gBW. The serial blood was collected from tail vein for 9 hours and was analyzed using spectrophotometer UV for unchanged paracetamol. The pharmacokinetic parameters of paracetamol were calculated  by recidual method and were analyzed by One Way ANOVA using 95% confidence interval. The difference between groups were analyzed using Post Hoc LSD-test. The results showed that the group II(paracetamol-mangosteen 1) and group III(paracetamol-mangosteen 2) increased of ka, Cpmaks, tmaks, t½ab, ke, t½el, CL,  AUC and decreased of ka, ke, and CL. The research showed that mangosteen juice doses 2 give higher decreased metabolism and decreased elimination of paracetamol in rat.

UJI AKTIVITAS PENGANGKAPAN RADIKAL 2,2-DIFENIL-1-PIKRILHIDRAZIL (DPPH) PIGMENT BAKTERIAL SIMBION DARI SARGASSUM POLYCYTRUM

Carotenoids are pigments that are found in plants, animals, microorganisms and humans. The pigment has many functions, one of which is as an antioxidant. There have been many studies of carotenoids in plants, but the types of carotenoids in bacteria is very rarely observed when the potential is not lost. The ability of marine bacteria is well known to produce carotenoids. Bacterial symbionts associated with Sargassum polycytrum can produce neoxanthin. This research will test the antioxidant activity by DPPH method on bacterial symbionts pigment extracts as compared with extracts of S. polycytrum. Based on the results obtained IC50 of 3.01 ± 2350 ppm was obtain for extract pigments S. polycytrum, 645 ± 2.11 ppm for pigment extract bacterial symbionts, and 564 ± 2.26 ppm for β-carotene as a comparison.

FAKTOR-FAKTOR YANG MEMPENGARUHI KEPATUHAN DOKTER TERHADAP PERESEPAN OBAT GENERIK DI INSTALASI FARMASI RSUD dr. R. GOETENG TAROENADIBRATA PURBALINGGA

Permenkes RI No. HK.02.02/Menkes/068/2010 dispute that  any mandatory to use generic drugs in public health facility. Recently, The Indonesian generic is have to using low, however the price is cheaper. The objective of this research to is  determinate the pecentage of doctor’s obedience to generic prescription and know the influance of knowladge, trust, generic drugs avaluable and patient’s influance to the obedience of docter’s to prescribe generic drugs in  Farmacy  Instalation of RSUD dr. R. Goeteng Taroenadibrata Purbalingga. The research was observational research, used cross sectional approach and retrospective data. The instrument of research is quisioner. Validity test used to know validitas of quisioner. Statistic analysis are used bivariat  analysis with chi square test and multivariat analysis used regresion logistic test. The result showed the percentage of doctor’ obedient was 58.3% and the unobedience 41.7%. Knowladge factor, trust, avaluability of drugs and patient influance each of them had significant influance to the level of doctor’s obedience to used generic drug’s. P value < 0.05. Factors of Knowladge, trust, avaluable of drugs and patient influance simultan had significant influance to the obedience of doctor. The conclution of this research is the persentage of doctor who obedience with generic prescribing were 58.3%.  The other side  the pertsentage of doctor who inobedience with generic prescribing were 41.7%. The higher of knowladge, trust, generic drugs avaluable and patient’s influance, so the higher of obedience of doctor in generic prescribe.

KARAKTERISASI MORFOLOGI SEL REKOMBINAN DAN PIGMEN HASIL FUSI PROTOPLAS INTERSPESIFIK Phaffia rhodozyma DAN Chlorella pyrenoidosaH Chick

Protoplast fusion technique is a process offusion between protoplasts of theorganism to other organisms, so that the resulting recombinant cells with varying character derived from its parent. This study did Phaffia rhodozyma interspesies fusion and Chlorella pyrenoidosa. The purpose of this study was to analyze the morphological characteristics of cells results protoplast fusion recombinant yeast P. rhodozyma and microalgae C. pyrenoidosa. Protoplast fusion technique performed in 4 phases, namely the isolation of protoplasts, protoplast fusion, and analysis of recombinant cell morphological characters. The results showed that the recombinant cells have morphological characters derived from the parent P. rhodozyma and of the parent C. pyrenoidosa. But was character of the parent P. rhodozyma more dominant. Recombinant cells capable of producing pigment and lutein astaksanthin. Astaksanthin pigment levels in recombinant cell height in a shorter time than the parent P. rodozyma.

PERBANDINGAN KADAR Cu (Tembaga) PADA KERANG DARAH (Anadara granosa) SEGAR DAN REBUS YANG DIJUAL DI PASAR BANJARSARI PEKALONGAN

Protein is a nutrient needed by humans. Marine animals is a provider of high-protein, one of the much-loved marine animals is Shell Blood. In addition to have a high content of protein, shells also have disadvantages because heavy metals can accumulate in the body, such as Cu metal. Cooking such as boiling process is expected to eliminate the toxic effects of the Cu content in the Shell Blood. In this study, samples were taken from Anadara granosa sold in markets Banjarsari Pekalongan. The method used is the Tampak spectrophotometry method, by reacting Cu with Sodium diethyltricarbonate to form complex compounds.
The results of this study are: the stability of the complex 15-minute period, the maximum wavelength of 440 nm, with a regression equation: y = 0.0987 x + 0.0756 and the correlation coefficient (r) was 0.9956, and obtained blood levels of Cu in the clam fresh is 8.75 ppm, 5.95 ppm and 19.36 ppm, and after boiling for 15 min levels down to 5.39 ppm, 4.47 ppm and 15.64 ppm. Boiling is effective in lowering levels of Cu by 16-45%.

FORMULASI DAN EVALUASI STABILITAS FISIK SUSPENSI IBUPROFEN DENGAN MENGGUNAKAN NATROSOL HBr SEBAGAI BAHAN PENSUSPENSI

Ibuprofen it a non-steroidal anti-inflammatory drug which is practically insoluble in water it can be made in the form of suspension. Suspension requires suspending agent  such as natrosol HBR for increasing the viscosity and reducing the rate of sedimentation that can produce stable suspension. The aimed of this research is to find the influence of natrosol HBR as the suspending agent toward physical stability of suspension. The method used in the manufacture of ibuprofen suspension is dispersion method. Ibuprofen suspensions were made with variation concentrations of 1%; 1.5%; 2% natrosol HBR respectively. In order to know the physical stability of suspension, it should be carried out organoleptic, density test, particle size distribution, viscosity test, sedimentation volume, redispersibility and pH test. The test result for three formulation suspensions for 30 days storage showed that the physical stablility changed in various showed concentration of natrosol. This research shown that the formulation with concentration of Natrosol Hbr 1% showed the best physical stability of the three formulas which indicated by the required results of the density test, sedimentation volume and redispersibility as well as the best particle size distribution and viscosity of the three formulas ibuprofen suspensions.
